Lupin "accidentally" forgot to take his potion, also disobeying 
Dumbledore's wishes, and endangering the whole school. 
It's only then that Snape "accidentally" lets slip that Lupin is a 
werewolf. 
Snape probably felt that his silence was conditional on 
Lupin holding up his end of the deal, and anyway, it was an 
accident, right?  As a bonus, it also gives credence to the threat 
Snape makes in GoF,  that he just might "slip" veritaserum into 
Harry's pumpkin juice.
