 Slytherin has certainly theirs, the chamber of secrets, which can only be opened by one who speaks parseltongue.  
  
However, I think Hufflepuff's chamber could be equally selective in who it will let in. I don't think hard work is selective enough. Maybe a sequence of tasks for which you have to show persistence to get them done.

a riddle or something like that - Hermione/Cho Chang could open that
