I suspect that Snape didn't know how much Quirrell knew. Snape
realized that Q was going after the stone, and probably assumed that
if he bothered trying in the first place, then he must know a way to
get through. As for Q -- maybe he didn't know about Fluffy, or maybe
he knew and was counting on Voldy for help which Voldy wasn't quite
strong enough to give yet. Or maybe he wasn't trying to steal the
stone at all, that first time; maybe it was a scouting expedition to
find out what the defenses were so he could be properly prepared for
the real theft attempt at the end of the book.
