OK, this baffled me too, so I went to get PoA, and here is page 278
UK.
Lupin threatens Peter and tells him not to move. They are outside the
Willow. The grounds were dark, with the only light from the castle.
"Silently they tramped through the grounds, the castle lights growing
slowly larger... And then --
A cloud shifted. There were suddenly dim shadows on the ground. Their
party was bathed in moonlight."
THEN Lupin changed.
He didn't change until the light hit him. The moon was behind a
cloud. It came out and he changed. So, by this logic, had the moon
stayed behind a cloud until they got to the castle, Lupin might not
have changed.
So, COULD he have stayed indoors in a windowless room??? That makes
no sense whatsoever. Someone help!

That's possible. Maybe it only has so long a "holding time" after
Snape makes it. He did tell Lupin to drink it right away earlier in
the book.
Pippin would tell us that Lupin saw the potion in Snape's robe while
examining him after the kids knocked him cold and ignored it. :)
