The Sergeant Majorette says

Snape knows who Lupin is. What he doesn't know is that the parchment 
is a map (until he goes to Lupin's office to bring him his wolfsbane, 
just before the big Shrieking Shack scene).

Snape tried several ways to find out what the parchment was, with no 
result until he told it his name ("I, Severus Snape, master of this 
school...")

Lupin's just messing with Snape. Everybody probably whispered about 
Snape being a vampire when he was in school, and how annoyingly 
ironic to realize that nobody (as far as he knew) had gotten the hint 
from the werewolf essay, but that the vampire essay would inevitably 
start up the whispering again about Snape himself. Remember the Drag!
Snape boggart? Lupin can be a real b****-- no, wait, let me rephrase 
that. Our Remus has some sharp fangs any time of the month.
