Lupin had tried to help – possibly call him a werewolf in front of
all the other students?

I don't think the Prank happened at that time, because James is still hexing
Snape, Lupin on the other hand was torn between his friends and his compassion
for Snape and the bullying that his friends were doing. Remember, this is
their fifth year and that was when they started to go with Lupin during the full
moon, until then, he had been alone. He is still unsure of their acceptance of
him.
