Perhaps the spiders were just in the castle.  So long as the chamber was
still closed, the serpent couldn't get to them.  Now that the chamber was
opened, the lives of the spiders were in danger.

Another possibility is that the spiders weren't so much afraid of being
eaten but afraid of some magical ability of the basilisk.
