IMO, she is now ill-disposed to Dumbledore because Fudge is. As long
as Fudge and Dumbledore were on friendly terms, Umbridge is no threat,
since she wants to be in the Minister's good grace. But now, that
Albus and Cornelius have broken, Umbridge became an enemie, because
she wanted to demonstrate Fudge her loyality. (Not that I think she is
really loyal to Fudge, but she wanted him to think so). But she went
further than Fudge, because now she is openly on his side, therefore
she knew that if Harry and Dumbledore turned out right in the end, her
power is threatened. Only as long as Fudge has the say, she is safe
(at least for that moment, I don't doubt that she would have used an
opportunity to overpower Fudge, if it were given to her). I don't
think she ever cared if Harry spoke the truth, she just wanted to make
sure nobody listens to him.
