I think the fact that JKR has said that we'd learn about why some people become ghosts and some don't could be a clue that one of the characters will die and come back as a ghost.  I mean, if she wouldn't comment further than to say that we would find out in a later book, then I would think that ghosts are going to be somewhat significant later.
To answer the second question:  no, I don't think Remus is happy.  Sure, he gets by and is always cheerful when we see him, but there must be times when the difficulties of his life become almost unbearable.  I think that's the reason that so many of the women on this list (myself included) are so fond of him.  We're all drawn to a tortured man, I suppose--at least in books and movies.
And now, I'd like to add to the first question.  Exactly which character(s), if any, do you think are most likely to become ghosts?  I think the most likely candidates, based on the happiness factor, are Remus and Sirius.  (I don't think I need to go into the reasons for Sirius's unhappiness).  Or even Snape, come to think of it.  Although if anything happens to any of them, I don't know what I'll do...
