Maybe Lupin is referring to the Wolfsbane Potion, which isn't a 'cure' so much as it making transformation bearable and less dangerous. I'm too lazy to actually schlep alllll the way over to my bookcase and look (hey, it's Sunday!) to bother checking, but I remember Lupin saying that the WP removes the rampaging beast element from the werewolf transformation and makes him harmless, so all he does for the few days of the full moon is sleep in his office.
If there is a *real* cure, maybe it's something spectacularly horrible, so horrible that Lupin would rather live with his lycanthropy than go through the torture of having it removed. And if that's the case... poor Lupin.
