JKR has said in interviews in response to questions regarding some of her backstories of characters (and I'm sorry I can't point specfic interviews, but such things are collected at http://www.accio-quote.org) that James's parents were very wealthy; that James was born late in their lives after several unsuccessful attempts to conceive, so they tended to dote him (which could make one a bit arrogant -- makes me think of Dudley a bit); and that they died natural deaths.  I don't recall anything she has said about Lily's parents though.
