I believe that comment was in response to the question of which house
JKR would be in, or like to be in. As far as I know, the only
comment by JKR regarding James being in Gryffindor was her lack of
contradiction when asked which position James played on the
Gryffindor Quidditch team.
As far as the OoP line, I wondered about that as well. I can't
decide whether it's definitive proof or not. It could be JKR's
subtle way of ending the controversy. Then again, as others
mentioned, it could simply be Harry's assumption.
